<title>Resumen</title>
<p>Las infecciones con el virus del dengue, transmitidas al humano por mosquitos del género <italic>Aedes </italic>sp., constituyen un problema de salud pública para las regiones tropicales y subtropicales del planeta. El ciclo de replicación del virus del dengue se lleva a cabo principalmente en el citoplasma de las células, en estrecha asociación con organelos membranosos como el retículo endoplasmático rugoso y el complejo de Golgi. En esta revisión se analiza la respuesta al estrés de estos organelos, ante la infección viral, tanto en células de vertebrados como de mosquitos, y se discuten las implicaciones de esta respuesta en la biología de estos agentes.</p>

<title>Abstract</title>
<p>Infections with dengue virus, transmitted to humans by mosquitoes of the <italic>Aedes</italic> sp. genus, constitute a public health problem for the tropical and subtropical regions of the planet. The replication cycle of dengue virus takes place mainly in the cytoplasm of cells, in close association with membranous organelles such as the rough endoplasmic reticulum and the Golgi apparatus. In this review, the stress response of these organelles, both in vertebrate and mosquito cells, to viral infection is analyzed and the implications of this response in the biology of these agents, discussed.</p>

<title><bold>Introducción</bold></title>
<p>El dengue es la enfermedad viral transmitida por mosquitos más importante dentro de la salud pública mundial. En la actualidad, más de 120 países, ubicados en las regiones tropicales y subtropicales del planeta, son endémicos para esta enfermedad, y se estima que cerca de dos tercios de la población del mundo vive en zonas de riesgo a la infección (<xref ref-type="bibr" rid="ref1">1</xref>,<xref ref-type="bibr" rid="ref2">2</xref>). El virus del dengue (DENV) es transmitido a los humanos por la picadura de mosquitos hembra del género Aedes, especialmente por <italic>Aedes aegypti</italic> en zonas urbanas. Aunque la infección por el DENV suele cursar de manera asintomática, la infección también puede presentarse con manifestaciones clínicas diversas, que van desde un dengue sin signos de alarma (fiebre por dengue) hasta un dengue grave (dengue hemorrágico) (<xref ref-type="bibr" rid="ref1">1</xref>). El dengue sin signos de alarma se caracteriza por fiebre alta (≥38 °C), cefalea, dolor retrocular, artralgias, mialgias, malestar general, entre otras manifestaciones, y suele resolverse en cuestión de días. Sin embargo, una fracción de los pacientes puede evolucionar a dengue grave, el cual se caracteriza por extravasación de plasma y hemorragias, que pueden llevar a un choque hipovolémico y a la muerte (<xref ref-type="bibr" rid="ref2">2</xref>,<xref ref-type="bibr" rid="ref3">3</xref>,<xref ref-type="bibr" rid="ref4">4</xref>). Los hallazgos de laboratorio en pacientes infectados incluyen trombocitopenia, hemoconcentración o alteraciones en los tiempos de coagulación y en la homeostasis de los lípidos (<xref ref-type="bibr" rid="ref4">4</xref>). Los síntomas del dengue pueden ser fácilmente confundidos con otras enfermedades virales, y aun con enfermedades bacterianas; por ello, es necesario el diagnóstico confirmatorio por laboratorio (<xref ref-type="bibr" rid="ref1">1</xref>,<xref ref-type="bibr" rid="ref2">2</xref>,<xref ref-type="bibr" rid="ref3">3</xref>,<xref ref-type="bibr" rid="ref4">4</xref>). En la actualidad no existen tratamientos antivirales específicos para dengue, y aunque existe una vacuna disponible, la misma presenta limitaciones en su uso (<xref ref-type="bibr" rid="ref5">5</xref>,<xref ref-type="bibr" rid="ref6">6</xref>,<xref ref-type="bibr" rid="ref7">7</xref>).</p>
<p>El DENV pertenece a la familia <italic>Flaviviridae</italic>, género Flavivirus (del latín <italic>flavus</italic>, que significa amarillo; así que es el virus de la fiebre amarilla, el virus tipo de la familia). El virión del DENV posee simetría icosaédrica (esférico) y está compuesto por tres proteínas estructurales (C: cápside; prM: premembrana, y E: envoltura) y además presenta una envoltura lipídica. El genoma está compuesto por una molécula de ARN de polaridad positiva (es decir, que presenta la misma polaridad que el ARNm de las células), de aproximadamente 10 Kb. Otros miembros del género <italic>Flavivirus</italic> son el virus del Zika (ZIKV), el virus del oeste del Nilo y el virus de la encefalitis japonesa, también trasmitidos por mosquitos. Se han identificado 4 serotipos del DENV (DENV1-DENV4), que están determinados por la variabilidad genética de la proteína E (<xref ref-type="bibr" rid="ref7">7</xref>,<xref ref-type="bibr" rid="ref8">8</xref>,<xref ref-type="bibr" rid="ref9">9</xref>). La infección del humano con un serotipo en particular confiere inmunidad prolongada o de por vida. Sin embargo, una persona puede volverse a infectar con un serotipo distinto y padecer dengue por segunda ocasión (<xref ref-type="bibr" rid="ref8">8</xref>,<xref ref-type="bibr" rid="ref9">9</xref>,<xref ref-type="bibr" rid="ref10">10</xref>). Además, las infecciones secundarias presentan riesgo aumentado (entre 100 y 1000 veces) de evolucionar a dengue grave. Las infecciones terciarias y cuaternarias, aunque en teoría posibles, ocurren solo en muy raras ocasiones, debido a la inmunidad cruzada que se genera después de la segunda infección (<xref ref-type="bibr" rid="ref9">9</xref>,<xref ref-type="bibr" rid="ref10">10</xref>).</p>

<sec>
<title><bold>Ciclo replicativo del virus del dengue</bold></title>
<p><italic>In vitro</italic>, el DENV muestra un amplio tropismo y es capaz de infectar a una gran diversidad de células que incluyen no solo células humanas y de mosquito, sino también de varias especies de animales, como monos y hámsteres (<xref ref-type="bibr" rid="ref7">7</xref>). El ciclo de replicación del DENV inicia con la interacción de la proteína E del virión con diversos receptores localizados en la membrana de las células diana como los glucosaminoglucanos, lectinas tipo C como DC-SING, receptores de manosa, receptores de fosfatidilserina o proteínas de la inmunoglobulina-mucina de linfocitos T (<xref ref-type="bibr" rid="ref11">11</xref>,<xref ref-type="bibr" rid="ref12">12</xref>). Una vez unido, el virión se internaliza a la célula por endocitosis mediada por clatrina y es transportado a endosomas tempranos o intermedios. En los endosomas, el bajo pH induce cambios conformacionales profundos en la proteína E que llevan a la fusión de la envoltura lipídica del virión con la membrana del organelo y a la subsecuente liberación de la nucleocápside al citoplasma celular. El desensamblaje de la cápside es un proceso poco conocido, que termina con la liberación del genoma viral en el citoplasma y su traducción a proteínas en los ribosomas del retículo endoplásmico rugoso (RER). Dada su naturaleza monocistrónica, el genoma es traducido como una poliproteína, procesada por la proteasa viral NS3 y proteasas del hospedero (algunas aún desconocidas), para generar las tres proteínas estructurales C, prM y E, y siete proteínas no estructurales (NS1, NS2A, NS2B, NS3, NS4A, NS4B y NS5). Estas últimas cumplen funciones durante la replicación viral y además modulan la respuesta innata de la célula (<xref ref-type="bibr" rid="ref12">12</xref>,<xref ref-type="bibr" rid="ref13">13</xref>,<xref ref-type="bibr" rid="ref14">14</xref>).</p>
<p>La replicación del genoma viral se lleva a cabo en invaginaciones de la membrana del RER, que el propio virus induce, conocidos como paquetes vesiculares o VP (<xref ref-type="bibr" rid="ref15">15</xref>). Dentro de los VP se encuentran las proteínas NS3 y NS5 formando un complejo. La proteína NS3 posee una función catalítica como proteasa, pero presenta también funciones de helicasa; por su parte, la proteína NS5 es la polimerasa viral ARN dependiente de ARN, y posee además actividad de metiltransferasa (<xref ref-type="bibr" rid="ref13">13</xref>,<xref ref-type="bibr" rid="ref14">14</xref>,<xref ref-type="bibr" rid="ref15">15</xref>). También asociada a los VP, pero por el lado del lumen del RER, se encuentra la proteína NS1, presumiblemente cumpliendo funciones de andamiaje para el complejo replicativo (<xref ref-type="bibr" rid="ref15">15</xref>,<xref ref-type="bibr" rid="ref16">16</xref>,<xref ref-type="bibr" rid="ref17">17</xref>). Finalmente, embebidas en la membrana del RER se encuentran las proteínas NS4A y NS4B, las cuales son responsables de inducir la curvatura de la membrana (<xref ref-type="bibr" rid="ref15">15</xref>,<xref ref-type="bibr" rid="ref18">18</xref>).</p>
<p>Una vez que el genoma ha sido replicado a través de intermediarios de replicación de ARN de doble cadena, el proceso de ensamblaje comienza con la asociación de dímeros de la proteína C con el ARN viral progenie, seguido de la gemación de membranas del RER que contienen el complejo glucoproteínico prM-E. Durante el ensamblaje y la liberación de las partículas nacientes, proteínas del hospedero regulan el tráfico intracelular y la adición y procesamiento de azúcares a la glucoproteína E. Posterior al ensamblaje, los viriones inmaduros se transportan por medio de la vía secretora clásica o constitutiva, a través del aparato de Golgi hacia el espacio extracelular. El último paso en la maduración del virión es un reordenamiento de prM-E debido al pH ácido del Golgi y a la acción de una proteasa residente de Golgi llamada <italic>furina</italic>. Las partículas virales se liberan de la célula mediante exocitosis (<xref ref-type="bibr" rid="ref14">14</xref>,<xref ref-type="bibr" rid="ref18">18</xref>,<xref ref-type="bibr" rid="ref19">19</xref>). La <xref ref-type="fig" rid="gf1">figura 1</xref> muestra un esquema del ciclo replicativo completo del DENV.</p>

La replicación del genoma viral y el ensamblaje ocurre en paquetes vesiculares en el retículo endoplasmático (5). Los viriones inmaduros se transportan por medio de la vía secretora constitutiva, a través del aparato de Golgi hacia el espacio extracelular donde ocurre la maduración final de las partículas virales qué ocurre gracias a la acción de la furina (6). Los viriones maduros son transportados mediante exosomas (7) hasta su liberación al exterior celular junto con la proteína NS1 hexamérica (8).</p>

<p>Junto a los viriones, las células infectadas con DENV también secretan NS1 a la glucoproteína no estructural. En el citoplasma de la célula, NS1 se encuentra asociada, como un dímero, a las membranas intracelulares y a los complejos de replicación (<xref ref-type="bibr" rid="ref16">16</xref>,<xref ref-type="bibr" rid="ref17">17</xref>,<xref ref-type="bibr" rid="ref20">20</xref>). Sin embargo, una fracción de NS1 es secretada de la célula como hexámero (<xref ref-type="bibr" rid="ref16">16</xref>,<xref ref-type="bibr" rid="ref17">17</xref>,<xref ref-type="bibr" rid="ref20">20</xref>,<xref ref-type="bibr" rid="ref21">21</xref>). En células de vertebrados, NS1 es secretada siguiendo la misma ruta clásica de secreción de los viriones, es decir, a través del complejo de Golgi (<xref ref-type="bibr" rid="ref21">21</xref>). En contraste, en células de mosquitos, NS1 es secretada a través de una ruta no clásica, independiente del aparato de Golgi y que depende de caveolina y otras proteínas asociadas (<xref ref-type="bibr" rid="ref22">22</xref>,<xref ref-type="bibr" rid="ref23">23</xref>).</p>

<sec>
<title><bold>Respuesta de estrés del retículo endoplasmático rugoso</bold></title>
<p>La respuesta de estrés del RER, también conocida como respuesta UPR (por sus siglas en inglés <italic>Unfolded Protein Response</italic>) ocurre como una respuesta de la célula al excederse la capacidad de procesamiento del RER, durante la síntesis de proteínas (figura 2). El exceso de proteínas mal plegadas o incorrectamente ensambladas es detectado por la célula a través de la proteína Bip (también conocida como GRP78), que actúa como sensora y bloqueadora de las tres vías de activación PERK, IRE-1 y ATF-6. La activación de la UPR a través de sus vías IRE-1 y ATF-6 conduce al aumento en la síntesis de lípidos y a la activación de genes de proteínas chaperonas; ello resulta en una expansión del volumen y en la capacidad de plegamiento del RER. Por su parte, la activación de la vía PERK induce la fosforilación del factor de iniciación eIF2α y el arresto general de la traducción de proteínas. Si la respuesta UPR no logra restablecer la homeostasis celular y la situación de estrés no se corrige, la célula termina activando cascadas de muerte celular programada (autofagia o apoptosis).</p>

<p><bold>A</bold>) Durante la infección del DENV, las células hospederas pierden la homeostasis celular, entre otras causas, por la acumulación de proteínas no o mal plegadas. La vía de estrés del retículo endoplasmático rugoso (UPR) tiene tres sensores (ATF6, IRE1 y PERK) que se encuentran unidos a la chaperona BiP (GRP78), la vía de UPR se activa cuando se desacopla la proteína BiP de los sensores debido a la presencia de proteínas mal plegadas. Las vías ATF6 e IRE1 conducen a la sobrexpresión de chaperonas y de contenido lipídico para aumentar la superficie de procesamiento del retículo endoplasmático. La activación de IRE1 también conduce a la activación de XBP1 que se transloca al núcleo para la activación de los genes ERAD. La vía PERK activa a eIF2α que bloquea la síntesis de proteínas hospederas. De no recuperar la homeostasis de la célula, la activación de la UPR puede culminar con la activación de autofagia o la apoptosis. <bold>B</bold>) En células infectadas con DENV se induce una respuesta de estrés del Golgi (GSR) a través de la activación de TEF3, el cual es desfosforilado y translocado al núcleo; allí activa la vía GASE para inducir la expresión de los genes <italic>GOLGA2</italic> y <italic>GCP60</italic>, entre otros.</p>

<p>Dada la estrecha relación entre la replicación de los flavivirus y el RER, no es de extrañar que la replicación del DENV active la respuesta de UPR en las células infectadas (<xref ref-type="bibr" rid="ref24">24</xref>,<xref ref-type="bibr" rid="ref25">25</xref>,<xref ref-type="bibr" rid="ref26">26</xref>). Sin embargo, además de activarla, el DENV es capaz de modular la UPR y utilizarla para generar una respuesta celular de supervivencia. La UPR tiene interacciones complejas con la maquinaria de autofagia celular, los mediadores de la apoptosis, la inmunidad innata y las reacciones proinflamatorias. Por lo tanto, el DENV modula selectivamente estos mecanismos celulares durante la infección para mejorar tanto la replicación viral como su capacidad de evadir la inmunidad innata del huésped (<xref ref-type="bibr" rid="ref25">25</xref>,<xref ref-type="bibr" rid="ref27">27</xref>).</p>
<p>La infección por DENV activa las tres vías de la UPR; la vía de PERK se activa temprano en la infección, seguida de la activación de la vía IRE1-XBP1 en la mitad de la infección y de la ATF6 más tardíamente en la infección. La activación de la vía PERK es parte de la respuesta antiviral de la célula, ya que resulta en atenuación de la traducción de proteínas. Sin embargo, durante la infección por DENV, la fosforilación de eIF2α dependiente de PERK solo se activa transitoriamente y temprano durante la infección (a las 6 horas posinfección, aproximadamente), y luego se revierte rápidamente (<xref ref-type="bibr" rid="ref27">27</xref>). De esta forma, la infección por DENV asegura la producción continua de sus proteínas y facilita la producción de viriones. Se desconoce el mecanismo de desfosforilación y activación del eIF2α, pero se ha sugerido que involucra la participación de proteínas del DENV (<xref ref-type="bibr" rid="ref25">25</xref>,<xref ref-type="bibr" rid="ref26">26</xref>,<xref ref-type="bibr" rid="ref27">27</xref>,<xref ref-type="bibr" rid="ref28">28</xref>,<xref ref-type="bibr" rid="ref29">29</xref>). La infección por DENV también activa la vía IRE1-XBP1 y un subconjunto de genes activados por XBP1, involucrados en la degradación de proteínas asociada al RER (o respuesta ERAD, de sus siglas en inglés, <italic>Endoplasmic Reticulum Associated protein Degradation</italic>), lo que alivia el estrés de RER. La activación de la vía XBP1 es inducida por las glucoproteínas E y NS1 y las proteínas hidrofóbicas pequeñas ancladas al RER (NS2A, NS2B y NS4B). Además, el DENV puede inhibir los mediadores de la apoptosis vía IRE1-XBP1, lo que da como resultado una mayor supervivencia celular y una mayor replicación viral (<xref ref-type="bibr" rid="ref25">25</xref>,<xref ref-type="bibr" rid="ref26">26</xref>,<xref ref-type="bibr" rid="ref27">27</xref>,<xref ref-type="bibr" rid="ref28">28</xref>,<xref ref-type="bibr" rid="ref29">29</xref>).</p>
<p>La autofagia es un proceso catabólico celular que promueve la formación de vacuolas autofágicas para degradar y reciclar los constituyentes celulares (<xref ref-type="bibr" rid="ref26">26</xref>,<xref ref-type="bibr" rid="ref29">29</xref>,<xref ref-type="bibr" rid="ref30">30</xref>). Desempeña un papel importante en la regulación del crecimiento y desarrollo celular y está implicada en ciertas patologías. La activación de esta respuesta celular promueve la eliminación de componentes virales y facilita la presentación de antígenos virales. Sin embargo, se sabe que el DENV modula el proceso, lo cual favorece la infección, ya que la inducción de autofagia protege a células infectadas de la apoptosis (<xref ref-type="bibr" rid="ref26">26</xref>,<xref ref-type="bibr" rid="ref31">31</xref>,<xref ref-type="bibr" rid="ref32">32</xref>,<xref ref-type="bibr" rid="ref33">33</xref>,<xref ref-type="bibr" rid="ref34">34</xref>). Se ha demostrado que la activación de la autofagia durante la infección por DENV requiere la activación de la vía PERK de la UPR (<xref ref-type="bibr" rid="ref32">32</xref>,<xref ref-type="bibr" rid="ref34">34</xref>,<xref ref-type="bibr" rid="ref35">35</xref>). La vía PERK activada regula la sobreproducción de especies reactivas de oxígeno e incrementa el reciclaje de la autofagia durante la infección por DENV. Aunque se considera que la contribución de las especies de oxígeno a la formación de autofagosomas es modesta, PERK cumple un papel importante en el mantenimiento de altos niveles de autofagia y contribuye a la producción de partículas virales maduras e infectivas. Además, las vesículas de doble membrana formadas durante la autofagia proporcionan una plataforma para la replicación viral. Por lo tanto, se ha sugerido que la UPR desempeña un papel importante en la mediación y activación de la autofagia en células infectadas por DENV, lo que facilita la replicación viral (<xref ref-type="bibr" rid="ref32">32</xref>,<xref ref-type="bibr" rid="ref33">33</xref>,<xref ref-type="bibr" rid="ref34">34</xref>,<xref ref-type="bibr" rid="ref35">35</xref>).</p>
</sec>
<sec>
<title><bold>Respuesta de estrés del aparato de Golgi</bold></title>
<p>El aparato de Golgi es un conjunto de membranas lamelares cuya principal función es recibir las proteínas recientemente sintetizadas en el retículo endoplásmico, procesarlas postraduccionalmente y transportarlas hasta diferentes organelos, así como a la membrana plasmática y al espacio extracelular; por ello, la función de este organelo es esencial para procesos celulares, como el metabolismo, la diferenciación, la proliferación y la migración celular (<xref ref-type="bibr" rid="ref36">36</xref>,<xref ref-type="bibr" rid="ref37">37</xref>,<xref ref-type="bibr" rid="ref38">38</xref>). El volumen del aparato de Golgi en el interior de la célula puede sufrir adaptaciones en respuesta a la demanda celular, sin que esto implique un desequilibrio homeostático. Sin embargo, cuando esta demanda sobrepasa la capacidad funcional del aparato de Golgi, las proteínas no son procesadas ni secretadas correctamente. Para compensar la sobrecarga funcional y evitar fallas en el procesamiento y secreción de proteínas, la célula activa varias vías de señalización que buscan restablecer la homeostasis del aparato de Golgi, lo cual se conoce como la <italic>respuesta de estrés del aparato de Golgi</italic> (GSR, por <italic>Golgi Stress Response</italic>) (<xref ref-type="bibr" rid="ref38">38</xref>,<xref ref-type="bibr" rid="ref39">39</xref>,<xref ref-type="bibr" rid="ref40">40</xref>), así como se ve en la <xref ref-type="fig" rid="gf2">figura 2</xref>.</p>
<p>El aparato de Golgi posee regiones o zonas funcionales que incluyen la red de cis-Golgi, cis-Golgi, Golgi medio, trans-Golgi y la red de trans-Golgi. Cada una de estas regiones está relacionada con funciones específicas de este organelo (<xref ref-type="bibr" rid="ref41">41</xref>). A diferencia de la respuesta UPR, los mecanismos de activación de la GSR no han sido del todo dilucidados, pero con base en las zonas funcionales mencionadas, hasta el presente se han identificado seis rutas de activación de la GSR en células de mamíferos (TFE3, HSP47, CREB3, ETS, PG y Mucina (<xref ref-type="bibr" rid="ref42">42</xref>,<xref ref-type="bibr" rid="ref43">43</xref>). De todas ellas, la vía TFE3 (<italic>Transcription Factor</italic> E3) es la más estudiada hasta el momento, ya que su activación es esencial para el mantenimiento de las funciones estructurales de N-glucosilación y de transporte vesicular en condiciones de estrés del aparato de Golgi (<xref ref-type="bibr" rid="ref42">42</xref>,<xref ref-type="bibr" rid="ref43">43</xref>,<xref ref-type="bibr" rid="ref44">44</xref>). En un estado fisiológicamente normal del aparato de Golgi, TFE3 esta fosforilado en la serina 108 y es retenido en el citoplasma. Sin embargo, en presencia de GSR TFE3 se desfosforila y se transloca al núcleo, para unirse al elemento responsivo de estrés del aparato de Golgi y así activar la transcripción de genes diana relacionados con la modificación enzimática (<italic>SIAT4A, SIAT10, FUT1, B3GAT2</italic>), las proteínas estructurales de Golgi (GM130, GCP60, giantina) y con el transporte vesicular (STX3A, WIPI49, RAB20) (<xref ref-type="bibr" rid="ref43">43</xref>,<xref ref-type="bibr" rid="ref44">44</xref>).</p>
<p>En un estudio publicado recientemente, descubrimos que las infecciones por los flavivirus DENV y ZIKV activan la GSR en células infectadas (<xref ref-type="bibr" rid="ref45">45</xref>). Este estudio constituye el primer ejemplo en la literatura de inducción de GSR por condiciones fisiológicas, y no utilizando tratamientos farmacológicos, como tradicionalmente se hace. Durante el ciclo de replicación, los DENV y ZIKV están en íntimo contacto con membranas celulares y requieren la participación de la maquinaria de la célula para la producción de partículas virales, y su posterior procesamiento y transporte desde el RER hasta el medio extraceluar, a través del aparato de Golgi (<xref ref-type="bibr" rid="ref46">46</xref>,<xref ref-type="bibr" rid="ref47">47</xref>,<xref ref-type="bibr" rid="ref48">48</xref>). Así mismo, la proteína NS1 es secretada en células de vertebrados a través del aparato de Golgi, donde sufre a su paso modificaciones en sus cadenas de carbohidratos. Todos estos procesos de transporte vesicular y modificación postraduccional en las células infectadas parecen generar una mayor demanda del aparato de Golgi, lo cual resulta en la GSR por activación de la vía TFE3 (<xref ref-type="bibr" rid="ref45">45</xref>).</p>
<p>Ensayos de microscopía confocal de células, tanto de vertebrados (BHK21 y Vero-E6) como de mosquito (C6/36), infectadas con DENV y ZIKV, mostraron la fragmentación del aparato de Golgi al progresar la infección, medida utilizando como marcador la proteína estructural de Golgi GM130. Así mismo, en células infectadas se observó la translocación del factor TFE3 desde el citoplasma al núcleo, lo cual indica que en células infectadas se activa la vía TFE3; la activación de la vía TFE3 se corroboró al observar la sobreexpresión de los genes <italic>GOLGA2</italic> y <italic>GCP60</italic>, dos de los genes que son regulados por TFE3 y que codifican para las proteínas GM130 y la proteína residente del Golgi GCP60, respectivamente. Finalmente, también se observó que la sola expresión de la proteína NS1 en células de vertebrados es suficiente para inducir una GSR. NS1 es una proteína que llega a alcanzar altas concentraciones (ng/ml) en el suero de los pacientes, lo cual es compatible con la idea de que NS1, por sí sola, ya sea capaz de sobresaturar al aparato de Golgi durante la infección. Es interesante hacer notar que la replicación de un virus altamente lítico, como lo es el calicivirus felino, y cuya replicación no involucra el aparato de Golgi, no induce la respuesta de GSR, lo cual indica que la activación GSR no es una respuesta inespecífica a cualquier replicación viral (<xref ref-type="bibr" rid="ref45">45</xref>).</p>
</sec>
<sec sec-type="conclusions">
<title><bold>Comentarios finales</bold></title>
<p>Dada la estrecha asociación que existe entre la replicación del DENV, ZIKV y otros flavivirus con el RER y el aparato de Golgi, así como la elevada cantidad de proteínas virales que se producen en una célula durante la infección, no es de extrañar que en células infectadas se generen respuestas de estrés en ambos organelos. La infección por DENV causa estrés en el RER y activación de la respuesta UPR, como respuesta del hospedero. La evidencia experimental apunta a que el DENV modula esta respuesta del huésped para mejorar la replicación viral, al inhibir la apoptosis, al activar la autofagia y al suprimir la respuesta inmune innatas (<xref ref-type="bibr" rid="ref35">35</xref>,<xref ref-type="bibr" rid="ref49">49</xref>,<xref ref-type="bibr" rid="ref50">50</xref>). Más aún, un reporte reciente indica que los autofagosomas generados en células infectadas por DENV son infecciosos, ya que contienen genomas completos del virus (<xref ref-type="bibr" rid="ref48">48</xref>). Sin embargo, si bien se ha reportado que la activación de autofagia favorece la infección por DENV (<xref ref-type="bibr" rid="ref49">49</xref>), también existen reportes contradictorios que señalan que la autofagia podría inhibir el ciclo de replicación (<xref ref-type="bibr" rid="ref50">50</xref>), dependiendo del tipo celular. Por lo tanto, todavía queda mucho por entender sobre cómo afectan a la replicación viral, la respuesta UPR y la activación de la autofagia.</p>
<p>En el caso de la GSR, es claro que la infección por DENV y ZIKV activan dicha respuesta; sin embargo, aún se desconoce qué efecto ejerce dicha respuesta sobre la replicación viral, la secreción de viriones y de NS1; es decir, si la GSR favorece o antagoniza la replicación viral. Experimentos en curso en nuestro laboratorio buscan contestar esta pregunta evaluando la replicación de dengue en células impedidas de iniciar la GSR. Tampoco está claro cómo la activación y la modulación de las vías de estrés se suman a la patogénesis de la enfermedad en humanos durante la infección por DENV o ZIKV. Se necesitan más estudios para comprender la naturaleza compleja de estas interacciones, lo que proporcionaría objetivos útiles para la terapia antiviral y ampliaría nuestro conocimiento sobre la patogénesis del DENV. Dada la naturaleza aguda del cuadro clínico de dengue, y a que en la actualidad no existen tratamientos antivirales específicos contra este virus, valdría la pena considerar la posibilidad de manipular farmacológicamente estas respuestas de estrés como posible tratamiento para la patogénesis del dengue.</p>
