Abstract

Aquaporin-3 (AQP3) is expressed in human placenta from the onset of gestation until term. It is involved in physiological processes of the trophoblast such as migration and apoptosis and in several clinical dysfunctions, being reduced in preeclamptic placentas.


Placental exosomes, extracellular vesicles that derive mainly from the syncytiotrophoblast, have been reported in circulation since the sixth week of gestation. They may represent trophoblast changes during pregnancy and have begun to be used for the study of placental pathologies.


Our objective was to study the presence of AQP3 mRNA and protein in exosomes from plasma of pregnant women before the 20th week of gestation.


The samples were obtained with the approval of the bioethics committee and the signing of the informed consent.


The exosome-enriched fractions were obtained by differential centrifugation and immunoselection by binding to anti-CD63 (exosomal marker) and the presence of AQP3 and placental alkaline phosphatase (PLAP, syncytiotrophoblast marker) were studied using qRT-PCR and western blot.


The results showed that the exosome-enriched fractions contain placental exosomes. AQP3 mRNA and protein were also detected in the fractions. Future studies will determine if AQP3 level may be useful as an indicator of placental function throughout pregnancy.
